Growth Hormone Secretagogue
Ipamorelin is a small lab-made peptide (a short chain of just five amino acids, the building blocks of proteins). In studies it is described as one of the first compounds that very selectively prompts the body to release growth hormone. It belongs to a group of compounds that mimic ghrelin (a natural body hormone often called the hunger hormone) and has been used as a research tool to study selective growth-hormone release.
Ipamorelin switches on the ghrelin receptor, known by the code name GHS-R1a (the docking point on cells that the hunger hormone ghrelin normally acts through). Turning it on is studied for prompting the pituitary gland (a small gland at the base of the brain) to release growth hormone, through a path separate from the GHRH pathway (the other main way growth hormone release is triggered) [1]. A key point reported in the first studies is how selective it is: it prompted growth-hormone release without meaningfully raising other hormones such as ACTH, cortisol (the stress hormone), or prolactin, even at doses well above what was needed for the growth-hormone effect [1].
In awake animals, ipamorelin was reported to prompt a clear, dose-related release of growth hormone, and researchers have used it to compare growth-hormone release driven by the ghrelin receptor with release driven by GHRH [1]. Because it is so selective, it is a common research tool for studying ghrelin-receptor effects on the growth hormone and IGF-1 system (IGF-1 is a growth signal triggered by growth hormone) without stress hormones getting in the way [1].
Animal studies have looked at compounds of this type for effects on measures such as body-weight gain and bone mineral, to study the downstream results of prompted growth-hormone release [1].
The supporting evidence is mostly from animal and mechanism studies.
